In this study verification was low, but abnormality observed in some roots that probably were related to agar solidification. All established techniques for the miniature rose micropropagtion use solid medium; one of the common commercial mediums that have been mainly used for rose miniature micropropagation is MS medium. The concentration of hormones in medium is very important for shoot multiplication. In order to study shoot multiplication of miniature rose in this experiment, different level of 6-benzylaminopurin (BAP: 0, 8,16, 24µM) with the fixed level of naphthalene acetic acid (NAA: 0.07µM) were used. The experimental design was complete randomized block with 20 replications. For the rooting different level of NAA (0.1, 0.4, 0.7, 1 µM) used with the fixed level of BAP (2µM) in complete randomized block design within 10 replications. For shoot multiplication, fresh weight, dry weight and multiplication ratio and in the rooting, root number, root length and root volume were measured. Results showed that in the high concentration of BAP (16, 24 µM) the rate of shoot multiplication was higher but in low level of Bap (8µm) multiplied shoots had more fresh and dry weight. After 4 week subculture, these shoots transferred to the rooting medium. Results of this experiment showed that, those roots produced in low concentration of NAA (0.4µM) were longer.
