Abstract

Australian grapevine viroid (AGVd), an apscaviroid of the family Pospiviroidae, was found in vineyards of southern Iran for the first time. It had a relatively wide host range and caused stunting, leaf deformation, mottling and vein clearing in experimental hosts upon mechanical inoculation of nucleic acid extracts or agroinfiltration of the viroid infectious cloned DNA. Compared with the type isolate, the Iranian isolate of AGVd (AGVd-Ir) had a different conformation with an extra loop in the pathogenicity domain of the secondary structure. Mutational analyses using infectious clone of AGVd-Ir showed that disruption of the extra loop decreased the replication efficiency of the viroid but had no effect on its movement in the plant.
