چکیده

Regenerative medicine and tissue engineering focus on replacing and restoring damaged cells, tissues, and organs as a new discipline in biomedical research, and for this purpose, three-dimensional (3D) scaffolds are used combination with sources of stem cells. Damages of skin, as a mechanical barrier for the body against external factors, lead to physiological imbalances, disabilities or even death. Skin tissue engineering is one of the newest therapies in which the selection and optimization of scaffolds are very important. Acellular dermal matrix (ADM) is widely used in skin tissue engineering, because it provides a natural microenvironment for cellular adhesion and proliferation, as well as angiogenesis, and even antimicrobial activities. In this study, different methods of decellularization were tested in order to find the optimized method in which decellularization is achieved with the lowest damage to the matrix. Evaluated protocols included use of NaCl in combination with Triton-X-100/SDS, trypsin with Triton X-100 and trypsin with NaOH. Histological studies, using different general and specified staining methods, indicated that use of trypsin with NaOH was the best method for preparing a suitable ADM. Different preparation methods of hADM could influence the applications of this scaffold in tissue engineering because of various effects of decellularization agents upon matrix components.
