Abstract
Abstract: Escherichia coli O157:H7 is an important human pathogen causing haemoragic colitis(HC), hemolytic-ureamic syndrom(HUS) and thrombotic thrombocytopenic purpura. A number of protocols for detection of Escherichia coli O157:H7 from food specimens have been proposed, but PCR based methods has a higher sensetivity and specificity. In this study number of 100 ground beef samples were collected randomly from beef markets during the june of 2004. Escherichia coli O157:H7 isolated from one out of 100 samples using culture method by enrichment in modified trypticase soy broth (mTSB) and then plating onto sorbitol Mac Cankey agar supplemented with cifixime and potassium tellurite(CT-SMAC), cosequntly PCR from suspected colonies which were non sorbitol fermenting(NSF),using primers specific for O157 and H7 antigens gene. This method of isolation is a rapid,sensetive and specific method for isolation of Escherichia coli O157: H7
