Abstract

Pattern recognition receptors (PRRs) are the main sensors of pathogen and danger signals in innate immunity. They are mainly expressed by macrophages and dendritic cells of different organs. Toll like receptors (TLRs) are the most studied and best characterized PPRs which are responsible for sensing pathogen associated molecular patterns (PAMP). The contribution of PRRs in cerebral inflammation induced by microbial infection, tissue damage and cancer has not extensively been addressed so far. In the present study, human glioblastoma cells line U87 MG was used as a model to investigate the expression of TLR2, TLR4 and CD14 transcript in these cells and peripheral blood mononuclear cells (PBMC) of healthy human individuals. Real-time quantitative PCR was applied to quantify and compare the expression level of these genes in U87 MG cell line and PBMC of healthy individuals. According to our results, TLR2, TLR4 and CD14 transcripts are expressed in human gliblastoma cells. However, a significant down-regulation of CD14 and slight up-regulation of TLR2 transcripts were observed in U87 MG cell line in comparison to PBMC of healthy individuals. The lower expression level of CD14 in these cells in comparison to PBMC might have a potential implication of CD14 mediated cerebral pathology.
