Abstract

Background and Aim:Collagen is a key component of the extracellular matrix (ECM) and it is widely used for biomedical and pharmaceutical applications. In this study we used fish swim bladder (FSB) as collagen source & Nano Mg (0.97) Ag(0.03) O an antimicrobial agent to prepare the appropriate antibacterial wound dressing with simple materials and study its effect on bacterial growth. Methods:After removing the outer layer of FSB and cutting it into small pieces, a combination of physical and chemical methods including snap freeze-thawing in liquid nitrogen, and treatment with sodium dodecyl sulfate (SDS1%) & (SDS 0.05) were performed to lyse the cell membrane and cellular components. Followingfixation of the tissue in Bouin’s solution and dehydration for histological study the tissue section staining with Hematoxylin&Eosin, Hematoxylin weigrete- picro indigo carmine, picro fuchsin is accomplished. Colloidal nano Mg(0.97) Ag(0.03)o prepared with deionized water as their soluble used in combination with (FSB) after 24 hours incubation in a 25 °C shaker. Staphylococcus aureus was chosen because of its presence in a wound to study its sensibility to the dressing with Growth curve method. Growth of S. aureus in the cultures containing the prepared wound dressing which weighted 1gr was determined in comparison with control where no wound dressing was present. Results: Histological study of FSB before and after de-cellularization with SDS 0.5% & 1% confirmed the removal of cells and tissue but collagen. However as de-cellularization with SDS1% was destroying some of collagen tissue thus we used 0.5% SDS for preparing wound dressing. The anti-bacterial study of FSB + nano Mg(0.97) Ag(0.03) O on S. aureus’ growth using standard growth curve methods showed a decrease in the growth of the bacterium in presence of wound dressing compared with control. Conclusion:According to our histological results FSB is the appropriate material for wound dressing because of its high quantity of collagen with appropriate porous for absorbing wound exude and ventilation. Furthermore these porous structures made it an appropriate case for a 3-D structure of the growth of cells, tissue engineering applications and adding other antimicrobial Nano sized material.this dressing are active against s.aureus which present in wound.
