Abstract

Introduction: Breast, lung, and prostate carcinomas are among the most prevalent malignancies worldwide. Epithelial-mesenchymal transition (EMT) is a crucial process that plays important roles in embryonic development, wound healing, and cancer progression. This process facilitates the transformation of epithelial cells into mesenchymal phenotypes, which increase cell motility and invasion. The process of EMT is controlled by transcription factors (TFs) belonging to SNAIL zinc finger family (SNAL1 and SNAIL2), TWIST basic helixloop-helix family (TWIST1 and TCF3), and zinc finger E-box binding homeobox proteins (ZEB1 and ZEB2). This study unravels the importance of ZEB, SNAIL, and TWIST TF families on the metastasis and survival of breast invasive carcinoma (BRCA), prostate adenocarcinoma (PRAD), and lung adenocarcinoma (LUAD). Methods: We used GEPIA2 (http://gepia2.cancer-pku.cn/#index), an extensive and powerful online tool, to analyze the expression pattern of ZEB1, ZEB2, SNAI1, SNAI2, TWIST1, and TCF3 in LUAD, PRAD, and BRCA. Through the standard processing pipeline, GEPIA2 provides comprehensive analysis and evaluates the correlation between gene expression and survival. Discussion and conclusion: Higher expression of TWIST1 was detected in tumor samples of prostate and lung in comparison with normal tissues. Quit unlike TWIST1, the expression of TCF3 in tumor samples was considerably higher than their normal counterparts across BRCA, LUAD, and PRAD. Regarding ZEB1, ZEB2, SNAI1, and SNAI2, overexpression was more obvious in normal samples than in tumor ones. We also evaluated the correlation between EMT-TFs and the survival rate of BRCA, LUAD, and PRAD patients. We found a negative correlation between SNAI1 and survival in BRCA. In addition, the expression of SNAI1, SNAI2, and TWIST1 was negatively correlated with survival in LUAD. Last but not least, TWIST1 and more considerably TCF3 were negatively correlated with survival in PRAD.
