An Optimized Degenerate Polymerase Chain Reaction (PCR) for the Detection of Chicken Infectious Anemia Virus (CIAV) in Commercial Poultry Vaccines For quality control of biologicals of veterinary use, the absence of extraneous agents needs to be certified. one of the Requirements for quality control of avian viral vaccines is to demonstrate freedom from extraneous and adventitious agents, including Chicken Infectious anemia virus (CIAV). Chicken Infectious Anemia Virus can cause anemia, immunosuppression, and mortality in young chickens. To date the methods to detect CIAV in biologicals are time consuming and laborious. In this assay, an optimized degenerate polymerase chain reaction (PCR) for the detection of chicken infectious anemia virus (CIAV) was developed. The optimum concentration of MgCl2 and pH was 1.0 Mm and 9.2 and glyserol was used as an additive. After spiking of an ILTV vaccine with DNA of CIAV, the sensitivity of the PCR assay or the limit of detection for the primers and test was 3.056×10-9 ng/μl. In the presence of different viruses, including IBV, NDV, IBDV and ILTV, the primers and the test was able to amplify CIAV.The test described here may provide a rapid, sensitive and specific method for the detection of contamination of poultry vaccines with CIAV, and may be applied for quality control of live and killed vaccines. However, to validate this assay it has to be employed on a vast number of vaccine samples, while it is compared with other conventional methods.
