Combinationofseveraldrugsisoftennecessaryespeciallyduringlong-termtherapy.Thecompetitive bindingdrugscancauseadecreaseintheamountofdrugboundtoproteinandincreasethebiological activefractionofthedrug.TheaimofthisstudyistoanalyzetheinteractionsofLomefloxacin(LMF) and Colchicine(COL)withhumanserumalbumin(HSA)andtoevaluatethemechanismof simultaneousbindingofLMFandCOLtoprotein.Fluorescenceanalysiswasusedtoestimatethe effect ofdrugsontheproteinfluorescenceandtodefinethebindingandquenchingpropertiesofdrugs- HSA complexes.ThebindingsitesforLMFandCOLwereidentifiedintertiarystructureofHSAwiththe use ofspectrofluorescenceanalysis.TheanalysisoffluorescencequenchingofHSAinthebinaryand ternarysystemsshowthatLMFdoesnotaffectthecomplexformedbetweenCOLandHSA.Onthe contrary,COLdecreasestheinteractionbetweenLMFandHSA.Theresultsofsynchronousfluorescence, resonancelightscatteringandcirculardichroismspectraofbinaryandternarysystemsshowthat bindingofLMFandCOLtoHSAcaninducemicro-environmentalandconformationalchangesinHSA. The simultaneouspresenceofLMFandCOLinbindingtoHSAshouldbetakenintoaccountinthemulti- drug therapy,andnecessityofusingamonitoringtherapyowningtothepossibleincreaseofthe uncontrolledtoxiceffects.MolecularmodelingofthepossiblebindingsitesofLMFandCOLinbinary and ternarysystemstoHSAconfirmsthespectroscopicresults.
