Lipase is a type of hydrolyzing enzymes expressed in organisms to hydrolyze lipids and their derivatives. Some thermophilic organisms use lipase to be fed from lipids and oily components of their living environment. In this study, we extracted a novel lipase from bacillus ferdowsicus which is the type of thermophilic. First bacteria had an overnight in the media culture contained nutrient broth and 3% olive oil for 33 hours in 42˚c. Then media was centrifuged for 15min in 16000 rpm and lipase was extracted by using Q-sepharose column. The enzyme has a molecular weight about 45 kD. The enzyme activity was determined by treating with its substrate Para-Nitro Phenol Acetate (PNPA) in 60˚c for 30min. The purified enzyme has a specificity for Para-Nitro Phenol Palmitate (PNPP) and PNPA respectively and has an optimal pH around 6.5 and optimal temperature of 55˚c.
