Abstract
Recent studies suggest that oxidative stress is important in the etiology of multiple sclerosis (MS) and that it contributes directly to central nervous system (CNS) damage. Free radicals are side production massively in MS and it has also been indicated that free radicals are required for the phagocytosis of myelin by macrophages. In recent years the use of bioactive peptides as potent antioxidant has drawn the attention of researchers. This study was to identify the antioxidative potentials and amino acid sequences of peptides derived from egg white ostrich proteins. In this study, the isolated egg white proteins were hydrolyzed by papain. Protein hydrolysates were fractionated through an ultrafiltration membrane system with a 3000 Da molecular weight, and then were purified using reversed-phase high performance liquid chromatography (RP-HPLC) methods to obtain antioxidant peptides. The peptide fractions were assessed for free radical scavenging effects and also were in another system characterized for their antioxidant activity by measuring their ability to inhibition of linoleic acid autoxidation. Among the twelve peptides fractions, two peptides were observed to be the most power for their antioxidant effect on 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and values were 63 and 76 percent. In addition the two peptides were exhibited to inhibitory activity of linoleic acid autoxidation and values were 66 and 69 percent. The results showed that the digestion of egg white ostrich protein by protease papain can be used to produce antioxidant peptides and it may be help to downfall risks resultant of MS.
