Lack of adequate information regarding genotypic diversity within populations of plant pathogenic fungi is one of the most critical factors limiting breeding programs. in this study the genetic diversity of 22 pathogenic isolates of Fusarium solani from Razavi and Northern Khorasan provinces and 6 isolates from Tehran, Ardabil and Hamadan provinces were determined. For this purpose, ITS4 and ITS5 primers for determined rDNA-ITS region and also EI1 and ISSR primers which have been designed on the basis of conserved sequences in introns at 3´ end of rDNA and repetitive conserved regions respectively, were used. The amplification of rDNA-ITS divided all isolates in to ITS1 and ITS2 groups. Digestion results of the mentioned region were subjected to HeaIII and EcoRI enzymes, showed the more efficiency of first enzyme in creating polymorphism. Clustering was done with UPGMA method in NTSYS computer program. Clustering analysis of EI1 and ISSR primers showed suitable polymorphism among isolates, but the results reported here pointed out that EI1primer as a more reliable method for detecting genetic differences between isolates. Combinatory analysis dendrogram at the 75% similarity level grouped all the isolates in 25 genetic clusters without any correlation to their geographical origin and pathogencity. This is the first investigation concerning molecular researches in F. solani of potato in Iran.
