Abstract

Objective: Chicken infectious anemia virus (CIAV), the only member of the genus Gyrovirus in the family Circoviridae, is the causative agent of chicken nfectious anemia and cell-mediated immunosuppression in infected chick. It is a small DNA virus with a circular, covalently linked, negative-sense single-stranded genome. The genome has 3 open reading frames encoding 3 viral proteins (VP1, VP2, VP3). In the present study, samples from commercial broiler chickens from liver, spleen and thymus from Northeast of Iran (Khorasan), were screened by PCR and in positive samples, the major part of VP1 in three samples were sequenced. Material and method: 120 samples from commercial broiler chickens from liver, spleen and thymus in Northeast Iran were screened by diagnostic primers and then in three positive samples, the major part of VP1 with CAV primers was processed respectively. The edited sequences, along with respected sequences, deposited in genetic databases, were compared by multiple alignment and phylogenetic analysis tree, using BioEdit software and clustal W program. For phylogenetic analysis blosum 30 matrix, neighbor-joining method with 1000 replicate for bootstrapping were employed. Result and conclusion: The 582 bp amplified fragments of VP1 gene from 3 field samples were sequenced and similarity and identity with sequences in genetic databases were defined. Based ondeduced amino acid sequences of VP1 in phylogenetic tree, three Iranian strains were clustered in two groups. Three Iranian strains were clustered along with non-cell culture-adapted strains, separately from conventional cell-culture-adapted vaccinal strains. It is the first molecular study of VP1 gene of Northeast Iran strains of CIAV amplified from viruses circulating in commercial broiler farms.
