Abstract

During field and greenhouse surveys in 2009-10, Sesame, Cucurbit and Cucumber plants showing symptoms resembling those caused by phytoplasmas were collected from Sabzevar, Khaf and Torbat Heidarye in Khorasan Razavi Province and Uromye in Azarbaijan Gharbi province. The causal agent was transmitted from diseased plant to periwinkle by dodder (Cuscuta campestris) in greenhouse. The symptoms of the diseased plants were consisted of floral virescence, phyllody, proliferation and sometimes yellowing, cracking of seed capsules, germination of seeds in capsules, and formation of dark exudates on the foliage. Polymerase Chain Reaction (PCR) test using phytoplasma-specific primer pair P1/P7 and nested PCR using R16F2n/R16R2 primer pair was used in which it amplified products of expected size (1.8 and 1.2 kbp, respectively) from symptomatic plants. Direct PCR product cloned using pTZ57R/T cloning kit. The ligation mixture was used to transform competent cells of Escherichia coli DH5α. A P1/P7 clone was sequenced by fluorescent automated sequencing system. The obtained sequence was deposited in the GenBank database and compared with other sequences by CLUSTALW program. Restriction fragment length polymorphism (RFLP) of nested PCR products (1.2 kbp), in silico RFLP and phylogenetic analyses of 16S rDNA (Lee et al.,1998) sequences indicated that Sesame, Cucurbit and cucumber phyllodies (Seph1, Cubph, Cuph1 and Cuph2 respectively) were 100% identical and showed closest similarity with Vinca Virescence Phytoplasma, members of subgroup 16SrVI-A. Whereas Sesame phyllody (Seph2) revealed closest homology with phytoplasmas of the Juniperus Occidentalis witches’broom Phytoplasma members of subgroup 16SrIX-E.
