Abstract
Bene hull (Pistacia atlantica) is a natural resource and it has noticeable antioxidant capacity. Regarding the environmental concerns arising from the use of organic solvents, subcritical water extraction as a new environmental friendly technique was used. To optimize bioactive compounds extraction by subcritical water, response surface methodology with central composite face centered design was applied for bene hull. Subcritical water extraction conditions optimization includes temperature (110 - 200 °C), process time (30-60 minutes), and water to bene hull ratio (10-50). The Polyphenolic compounds (mg gallic acid/100g substance), reducing power (mg/ml ) and DPPH free radical scavenging activity (mg/ml) were measured as dependent variables. Optimal extraction conditions to obtain the maximum antioxidant activity were achieved at 196.81 °C, 52.57 minutes and water to bene hull ratio of 43.6. In these conditions, polyphenolic content, reduction power according EC50 and DPPH free radical scavenging activity according EC50 prediction were 2284 mg gallic acid/100g bene hull, 0.2002 mg /ml and 0.6284 mg/ ml, respectively. The results demonstrated usefulness of SWE as a suitable method to obtain biological materials from bene hull.
