Abstract

Background and Aim: The presence of bacteria in a wound is the main cause of the delay in its healing .This study’s aim is to prepare the appropriate antibacterial wound dressing with simple materials and study its effect on bacterial growth. Collagen is a key component of the extracellular matrix (ECM) and it is widely used for biomedical and pharmaceutical applications owing to its cell attachment capabilities, excellent biocompatibility, biodegradability and weak antigenicity. In this study we used fish swim bladder (FSB) as collagen source and Aloe Vera as a medicinal plant with anti-inflammatory, antimicrobial, anti-diabetic and immune-boosting properties. Methods: After removing the outer layer of FSB and cutting it into small pieces, a combination of physical and chemical methods including snap freeze-thawing in liquid nitrogen, and treatment with sodium dodecyl sulfate (SDS1%) were performed to lyse the cell membrane and cellular components. After fixation of the tissue in Bouin’s solution and dehydration for histological study the tissue section staining with Hematoxylin&Eosin, Hematoxylin weigrete- picro indigo carmine, picro fuchsin is accomplished. The Aloe Vera gel was prepared with 2% DMSO. Aloe Vera gel is used in combination with FSB after 1 hour incubating in a 25 °C shaker for preparing the wound dressing. The prepared wound dressing is cut into disc-shaped slices with 3mm diameter for the disc diffusion test. The filter paper disc with 1% SDS is prepared as control. Two bacteria Staphylococcus aureus and Staphylococcus epidermidis were chosen because of their presence in a wound to study their sensibility to the dressing with disc diffusion assay. Results: Histological study of FSB before and after de-cellularization shows the removal of cells and tissue and the remaining of collagen. The anti-bacterial study of FSB on two mentioned bacteria shows 12mm inhibition zone in presence of aloe Vera for both bacteria and about 11.5 mm inhibition zone for staphylococcus aureus, and 11 mm inhibition zone for Staphylococcus epidermidis in absence of Aloe Vera. No zone of inhibition for disc with SDS1%. Conclusion: According to our histological results FSB is the appropriate material for wound dressing because of its high quantity of collagen with appropriate porous for absorbing wound exude and ventilation. Moreover, FSB has inherently antibacterial activity. Furthermore these porous structures made it an appropriate case for a 3-D structure of the growth of cells, tissue engineering applications and adding other antimicrobial Nano sized material. Aloe Vera in addition to its antibacterial activity wets the dry wound and accelerates the wound’s healing
