Kelussia odoratissima Mozaff. Commonly known as “karaf-Koohi” is an endangered endemic medicinal plant in Iran which is widely used in regional distributions for several diseases treatment. Anti-inflammatory, cardiovascular and cerebrovascular effects contributed to Ligustilide compound which is found in essential oil from different plant parts. Continuous collection of plant materials from nature has led to its extinction. The purpose of this study was to establish the cell suspension culture of Kelussia which could be used for large scale production of active compounds. Inducted callus from sterile seedling explants was incubated in MS liquid basal medium for suspension culture establishment then suspended cells were cultured on liquid MS and B5 media supplemented with different combined hormones (( 2mg/l) 2.4-D + (0.5 mg/l ) kin, (2mg/l) 2.4-D+ (0.5 mg/l) BA, (2mg/l) NAA + (0.5 mg/l) BA and (2mg/l) NAA+(0.5 mg/l) Kin)) for 5 weeks. Results showed that growth pattern of K. odoratissima cells is a general sigmoid curves and maximal cell growth was obtained when suspended cells were cultured in liquid MS basal medium supplemented with 2mg/l 2.4-D+0.5 mg/l kin.
