Abstract

The development of heterocyclic scaffolds is a fast emerging subject in medicinal chemistry. Among these, benzo-fused sulfurated seven-membered lactams are endowed with a large spectrum of biological activities with a wide range of interesting, physiological activities. For example, Benzothiazepines play a unique role in drug discovery programs, as they display a wide spectrum of biological activities such as antibacterial [1], antifeedant [2], analgesic [3] and anticonvulsant [4] In this study, 5-hydroxymethyl-6-methyl-1,2,3,4-tetrahydro-2,4-pyrimidinedione(1) was prepared according to the previous procedure and treated with POCl to obtainquantitatively2,4-dichloro-5-chloromethyl-6-methylpyrimidine(2) as precursors. The reaction of compound (2) with 2-Aminothiophenol and treatment with appropriate secondary aminesattained the corresponding heterocyclic system derivatives of 2-substituted-4-methyl5,11-dihydrobenzo[b]pyrimido[4,5-e][1,4]thiazepine in good yields. . 3
