Tissue engineeringis trying tomaketissueswith characteristicssimilar to thoseof normal tissue. Natural and syntheticscaffolds are kind ofscaffolds which are used in tissue engineering. The reason of using scaffolds isthat they play the function ofthe extracellular matrix. Since in the decellularization tissues andorgans derived biological scaffold, cellantigens areremoved andmany ofthe structural and functionalproteinsof the extracellular matrixare maintained, thesescaffoldsare used successfully in tissue engineering . decellularization process whichis removing the nuclear and cellular materials, and this process must with minimizing any injury to the morphology, and biological activity of the extracellular matrix. The purpose ofthis research iscreatingaappropriate methodfor decellularizingbovine inter-vertebraldiscandmaking three-dimensionalscaffold which is derived from that. Inthisstudy, after sampling and providing bovine inter-vertebraldiscfrom Mashhad slaughter decellularization using a variety ofphysical methods(snap freeze-thaw) and chemical( use of SDS at concentrationsof0.1, 0.3, 0.5 % in the three groups) is done. The result of this study shows that prepared scaffolds withSDS 0.3 % decellularization tissue which formdisc successfully. Therefore,this bioscaffold could be asuitable doss forinvestigating cellbehaviorsandprobably an appropriate modelfor initialinvestigationinfibrouscartilage tissue engineering.
