Introduction: Cannabinoids are considered to be the main biologically active constituents of the cannabis plant that reported have anticonvulsant effects. Here we assesse C-fiber elimination effect which induced by neonatal capsaicin intraperitoneally injection (50 mg/kg,i.p) and daily administration of hydro alcoholic extract of cannabis (50 mg/kg.i.p) on latency and duration of Seizures induced by PTZ in 20- days-old rats. Methods: Two –day-old pups were divided into experimental (N=7) and control groups (N=7) and (ip) treated with Capsaicin or nothing, respectively and daily administration of ethanolic extract of cannabis (50 mg/kg.i.p) on the fourth day for 6 days. C-fiber destructions were confirmed by Ammonium hydrochloride (wipe test) . On the day 20th, Seizures were induced with PTZ (50 mg/kg. i.p) and the latency of unset and duration of generalized epileptiform behaviors was recorded. Results: experimental group shows significant difference in latency and duration of seizures induced by PTZ in comparison with control group. (P< 0.05) Conclusion: Degeneration of sensory neuron terminals by Capsaicin, lead to functional alterations of substrates at cortical levels. Cannabinoid receptor (CB1) stimulation by cannabis extract in brain has been overcome on these changes and reduces the severity of seizures.
