Introduction: Skin acts as a physical barrier between body and outside environmentand in case of injuries it is essential to cover the body with anappropriate dressing. In this regard, biological dressings have received special attention which is due to their specific properties it protect and close the wounds; relieve pain, maintain microbial control and areeconomically advantageous. One of the biological dressings is amniotic membrane (AM). AM is the inner layer of the amniotic sacand consists of three layers: one epithelial cell layer, a thick basement membrane and an avascular stroma. AM has been applied in a variety of clinical research such as repair burnsand Varicoseulcers, tissue reconstruction of vagina and urinary bladder and in ophthalmology for conjunctival reconstruction. Methods: In this study, human amniotic membranes were obtained from Mashhad branch of Royan Cord Blood Bank and 0.5 M NaOH was used to remove amniotic cells from the membrane. Mesenchymal stem cells were obtained from adipose tissue (Ad-MSCs) and after characterization; were seeded on denuded AM scaffold at a seeding density of 5 × 104 cells/cm2 and cultured for 21 days. To investigate attachment and viability of Ad-MSCs on scaffolds, tissues were fixed for histological staining at days 7, 14 and 21. Results: scanning electron microscopy and histological analyses revealed complete removal of cells after NaOH treatmentfrom AM. Human Ad-MSCsproliferated andproducedthree layers of cells on thescaffold.This shows that the AM is a suitable scaffold for culture of Ad-MSCs as it supports their attachment and can be used as a biological dressing in regenerative studies.
