Bean common mosaic virus (BCMV) is one of the most destructive and common viruses which infect common beans (Phaseolus vulgaris L.) throughout the world and cause mosaic symptoms and serious crop losses. In order to study the genetic variation of BCMV in north of Iran, a total of 100 leaf samples showing virus symptoms were collected from bean fields in Mazandaran and Golestan provinces during the growing season of 2014. Total RNA was extracted from all samples and used in RT-PCR analysis. RT-PCR using specific primers amplifies 860 bp fragments corresponding to the BCMV coat protein (CP) gene in 48 plant samples. Six PCR fragments were cloned into the pTG19-T vector and sequenced. Phylogenetic tree based on multiple sequence alignment of 860 nt of CP gene divided all BCMV isolates into 3 groups (I, II, III). Members of each group were divided into two subgroups: A, B. Mazandaran isolates were classified in the group IB along with two isolates from USA, while Golestan isolates were classified in the group IA. Phylogenetic analysis showed that Iranian isolates shared 87.9-98.6% nucleotide sequence identity with other BCMV isolates recorded in GenBank. Alignment of consensus sequences of these isolates with other isolates of BCMV available in GenBank showed Mazandaran isolates shared the highest (96-96.9%) and the lowest (87.9-88.3%) nucleotide sequence identity with RU1M isolate (USA) and Habin1 (South Korea), respectively; while Golestan isolates displayed the highest (98.3-98.6%) nucleotide sequence identity with NL1 (USA), and the lowest (88.5-88.7%) with Habin1. Mazandaran isolates shared 91.4% nucleotide sequence identity with Golestan isolates.
