Title : ( An in-silico screening on thermal stability of a series of bacteria dioxygenase for probiotic development )

Abstract

Background and aim: Industrialization of the societies leads to critical change in diet and environment. This transition led to appearance new compounds such as polycyclic aromatic hydrocarbons (RAH) in the human environments that are the essential causes of many diseases such as cancers. So, removal such pollutants from the environment and diet provide promising approaches for cancer prevention which are considered in this study through protein screening in order to probiotic development. Material and Methods: In this regard, a profile of the bacterial dioxygenases was collected based on literature. Corresponding nucleotide and protein sequences of the selected enzyme were retrieved from NCBI and UNIPROT databases. CD Search, Motif Scan, Blast, MEGA6, Hex and ClusPro2.0 programs were performed to evaluate data from literature. The thermal stability of the selected enzymes and domains were carried out using GROMACS5.07 under the OPLS force field at during 500ps for the NVT and NPT, and 20ns for MD stage. Results and Conclusion: The results of this study provide a series of catalytic domains that have already mentioned in the literature with degradation capacity of RAH. Moreover, the assessment of thermal stability of the selected domains and enzyme led to reveal a range of stability of them in different temperature. Conclusion: In general, this analysis led to present a series of temperature resistance enzyme, with the ability to RAH degradation, which could be applying for probiotic development based on genetic engineering.
