Cervical cancer is the third most frequent cancer in females worldwide, which if not detected and treated in time will lead to death. Auraptene a prenyloxycoumarin from Citrus species has been identified to have antitumor activity in breast, skin, colon, and prostate cancer cells. In this study, the cytotoxic and anticancer effects of 7-geranyloxycoumarin (auraptene), 6-geranyloxycoumarin (6-GC) and 5-geranyloxycoumarin (5-GC) were determined on HeLa (cervical cancer) and HDF (human dermal fibroblast) cells by MTT assay. The results of MTT assay revealed that the IC50 values of these compounds on HeLa cells were as follows: Auraptene (531, 245 and 151µg/ml), 6-geranyloxycoumarin (68, 53 and 23 µg/ml) and 5-geranyloxycoumarin (102, 25 and 21 µg/ml) after 24, 48 and 72h, respectively. Furthermore, The IC50 values of 6-GC and 5-GC were determined to be (79, 78 and 51 µg/ml) and(267, 140 and 133 µg/ml) after 24, 48, 72h, respectively on HDF cells. All in all, our results indicate that 6-GC and 5-GC can be presented as potential anti-cancer agents. Moreover, position of prenyl group may play a major role in the coumarin backbone cytotoxicity in cervical cancer.
