Abstract
Introduction: Staphylococcus epidermidis, the main pathogen in implant-related infections, is common bacteria of human skin that form biofilms on the surface of surgical implants. Staphylococcus aureus is a gram positive, commensal bacterium that colonize the human skin, nasopharynx and gastrointestinal tract which causes a variety of localized and invasive infections such as bacteremia, osteomyelitis, arthritis and prosthetic joint infections. Juniperus (FIG.1) genus has 6 species in Iran and is used in traditional medicine for wound healing, diabetes, tuberculosis, pneumonia and gastric ulcer. Geranium (FIG.2) genus is used as an antidiarrheal, anti-inflammatory and antidiabetic. The aim of this study was to investigate the possible antibacterial properties of leaf extract of these plants. Material and methods: methanolic extracts (FIG.3) of leaf freeze-dried powder were prepared by Perculation method (FIG.4) and then serial diluted extracts from 0.19 to 100 mg/ml concentrations were provided in distilled water and DMSO 5%. According to CLSI standards, 0.5 McFarland standard were created from overnight culture and diluted to 1:20 ratio. Serial dilutions of extracts with Muller Hinton Broth (MHB) media were loaded in 96 well microplates (FIG.5). Positive and negative control wells loaded with 100 µl media contained bacteria and media itself respectively. Minimum Inhibition Concentration (MIC) was investigated by TTC dye after 24 hours incubation. Results: the outcomes revealed that the MICs of both extracts had the same results for S. aureus and S. epidemidis that was 1.57 mg/ml and 12.5 mg/ml respectively. The Minimum Bactericidal Concentration (MBC) of Geranium extract for S. aureus and S. epidemidis was obtained 12.5 mg/ml and 25 mg/ml respectively, and for the Juniperus extract was obtained 25 mg/ml for both bacteria (Table 1). Discussion: Juniperus species are used for the treatment of hyperglycemia, tuberculosis, bronchitis, pneumonia, ulcers, intestinal worms, to heal wounds and cure liver diseases. The boiled leaves or fruits with animal fixed oil of J. Polycarpos are used for the treatment of earache in Hormozgan province, south of Iran. The phenol compounds, especially the flavonoids from Geranium spp. were reported to exhibit antiviral, antitumor, hepatoprotective, anti-inflammatory, anticancer and immune stimulant effects. according to this study, both leaf extracts demonstrated high bacteriostatic and bacteriocide properties which could be great candidates for staphylococcal infections treatment.
