Abstract

Osteoporosis, a progressive silent bone disease, affects 200 million people worldwide. The femoral necks and distal radius are one of the most common sites of fragility caused by osteoporosis. Therefore, functional solutions for the treatment of osteo- porotic fracture can be beneficial to a large population. The project aimed to develop a printable hydrogel (ink) for the construction of customized 3D scaffolds for healing osteoporosis-related fractures. Chitosan, as a biocompatible and osteoinductive polymer, was chosen as the basic component of the ink. The natural bone matrix was mimicked by adding nanohydroxyapatite. Alendronate was added to the hydrogel as osteoporosis first line drug treatment. To favor printability and shape fidelity, hydrox- yethylcellulose was added to enhance viscosity and introduce additional crosslinking. To overcome the issues with chitosan pH-dependent solubility, the inks containing quaternized chitosan were also tested. Ink composition and printing conditions were adjusted to obtain high-shape fidelity scaffolds. The biocompatibility of the printed material systems was tested in cell culture studies with MG63 cells. In future we will test the osteogenic potential of the proposed ink with the use of adipose-derived stem cells. The attempts to mix the ink with cells to form bioink will be also made
