Direct cell-to-cell communication is a vital requirement for the development, tissue regeneration, and maintenance of the natural physiology of multicellular organisms. Tunnel nanotubes exist as a cytoplasmic bridge between two near or far cells. They form cleft junctions between connected cells and mediate the exchange of cytoplasmic proteins, cellular organelles, lipids, and several other components. Recently, TNT formation has been shown to play a major role in the pathogenesis of many diseases. Viral particles, prions, fungal spores, organelles and other molecules can be transmitted by TNTs. The importance of TNTs goes beyond viral infections and cancer because they are involved in the pathogenesis of many neurodegenerative diseases. There is evidence that diseases such as Parkinson\\\\\\\\\\\\\\\'s, Huntington\\\\\\\\\\\\\\\'s and Alzheimer\\\\\\\\\\\\\\\'s disease can be caused by the passage of defective proteins through the TNTs :A preliminary search of the databases yielded a total of 10 articles, of which 5 articles were selected as source documents for this study. In neurodegenerative diseases such as Alzheimer\\\\\\\\\\\\\\\'s and Parkinson\\\\\\\\\\\\\\\'s, TNTs spread the disease by spreading alpha-synuclein fibrils and tau proteins to healthy cells, such as by spreading viral particles. Involvement of TNT in various pathologies makes it a goal in the treatment of various diseases ranging from cancer to dementia. However, because TNTs are primarily composed of actin, which is essential for various cellular processes and the maintenance of the cytoskeleton, balance must be maintained when targeting TNTs. Therefore, therapies should be modified in such a way that they affect only the pathological processes and maintain their normal function.
