Abstract Introduction: Myostatin gene or growth and differentiation factor 8 (GDF8) is one of the genes whose effect on growth has been determined. By coding a protein with the same name, this gene prevents the growth and differentiation of muscle fibers, especially during the embryonic period. If this gene can be deactivated or its activity reduced, it can cause higher growth of skeletal muscles. So far, many natural mutations have been identified in various animals, including cows, sheep, mice, etc., which have caused an increase in the growth of muscle mass (double muscle). In this study, the exon 1 and 5 of myostatin gene was genotyped in Boer and Gonabadi goats and Boer × Gonabadi hybrids using HRM method. Materials and Methods: Blood sampling was done from 20 Boer, 25 crossbred and 5 Gonabadi goats. After DNA extraction, a 378 bp fragment was amplified using a pair of specific primers that included terms of presence and type of mutation, PCR products of seven individuals were sequenced. Then, they were used as positive controls to determine the genotypes by HRM method. Results and discussion: After matching the sequencing results of the positive control samples with the melting curves of different individuals, it was found that the samples had three genotypes: AADD, AACD and ABDD. Conclusion: This study showed that HRM, compared to PCR-RFLP, PCR-SSCP and direct sequencing methods, is a less expensive, accurate and quick method for genotyping.
