Abstract

With increasing burden of some incurable diseases, tissue engineering has emerged as a new strategy in regenerative medicine. Using proper scaffolds, a good source of cells and specific growth factors can lead to production of desired tissues or organs to treat some problems or replace the damaged tissues in the body. Finding the best protocol for producing the scaffolds is very important in order to preserve the extracellular matrix, integrity and mechanical strength of the original tissue while removing all cells and antigens, which can elicit immune response in the recipient organisms. Moreover, addition of some nanoparticels or extracellular vesicles from different sources can improve the suitability of such scaffolds for various applications. In my lab we have focused on making natural scaffolds from various tissues and organs including skin, kidney, pericardium, fascia and so on, in animals and human, studied their properties, biocompatibility and suitability for cell growth, and have tried some of them in animal models. Here I present some of the results on protocols used for producing the scaffolds, their characterization, recellularization and also discuss the way forward to the clinic. Considering the need for such products in various surgeries, mass production of these scaffolds can overcome some problems in the clinic and pave the way from bench to bedside.
