Abstract
Lead is a highly toxic heavy metal, which can contaminate the bio-ecosystems via numerous routes. It can adversely affect many cells, organs and systems such as skeletal system. Bones are long-term repository for lead, containing approximately 90% of the total body burden in mammals and birds. Pb+ can readily replace Ca2+, leading to alteration in hormonal regulation of the calcium and osteoblast functions, resulting in osteoporosis due to bone depletion of Ca2+ ions. Herein, we have introduced a newly developed pharmacological agent, Poniost (arbitrary name) assessing its potential effects during experimental long-term lead poisoning (1000 ppm for 35 days in drinking water) using radiographic evaluation of bone density. The results have indicated that besides the lack of statistical effects of lead in changing the bone density compared to control littermates, oral administration of the drug (100mg/kg BW, bid, PO, n=8) in lead poisoned rats can significantly increase the density of spinous processes of the lumbar vertebrae (L1-L5) compared to control (water administrated; n=8) and lead poisoned (n=8) groups (p≤0.05). These results have indicated the potential bone protecting effects of this newly developed drug in augmenting the bone density and reducing the potential bone-depleting effects of environmental contaminant such lead, opening a new area of clinical use of this drug
