Abstract

Objective: Ostrich production has expanded considerably in Iran in recent years. Newcastle disease is a contagious and fatal viral disease that affects all species of birds including ratites. Three pathotypes of NDV have been recognized. Cleavability of the precursor F0 to F1 and F2 by specific cellular protease is the major determinant for virulence. The sequencing of the cleavage site of the F gene has provided researchers with a valuable tool for differentiation of virulent from low virulent and avirulent patotypes. In a past few months high mortalities appeared in ostrich flocks of Noertheast Iran. In this study involvement of vvNDV strains in ostrich flocks were investigated. Material and Methods: Clinical and macroscopic signs of vvNDV were observed in many A numbaffected flocks of Khorasan Razavi province, located in Northern-East part of Iran submitted for diagnosis. For virus isolation, brain tissues from a number of flocks were collected and homogenized. The brain homogenates were inoculated into allantoic fluid of embryonated chicken eggs, using standard protocols. The allantoic fluids were examined by hemagglutination (HA) and hemagglutination inhibition (HI) assays with NDV-specific antibodies. NDV positive allantoic fluids were processed for RNA extraction, followed by RT-PCR assay using specific primers which were capable of amplifying the cleavage site of F gene. The PCR products of two samples were sequenced and subjected to nucleotide and amino acid analysis. Results and Conclusion: The isolates from young ostrich flocks were all positive in HA and HI assays and RT-PCR examination. Sequence analysis of two PCR products confirmed that the isolates were virulent NDV strains. They possessed amino acid sequence motif of 111-GRRQKRF-117 in their F0 cleavage site, characteristics of virulent strains. According to this investigation NDV, circulating in recent outbreak of ND in ostrich flocks of Northeast Iran, are virulent and identical to the virus isolated from broiler and laying chicken flocks in the same region.
