Hemagglutinin9 gene (PCAG-sH9) alone, fused to 4 or 8 repeats of 29 amino acids (P 29) from C3d coding sequence (PCAG- sH9+4/8P29C3d), fused to Fc part of IgY coding sequence (PCAG-sH9+FcIgY) or fused to 8P29C3d and FcIgY coding sequence (PCAG-sH9+8P29C3d +FcIgY) DNA vaccines were tested in chickens. Expression of DNA vaccine constructs were confirmed by culture media and transfected cells with DNA vaccine plasmids by ELISA and immunofluorescence assays. The vaccines were injected intramuscularly to H9 free groups of 9 chickens each at 8 and 11 weeks of age. Anti-H9 antibody was measured by Hemagglutination Inhibition (HI) assay. Results suggested that fusing Fc IgY gene to PCAG-sH9 did not enhance immunogenicity of H9 DNA vaccine. In contrast, when 8P29C3d was fused and PC AG-sH9+8 P29C3d+ Fc IgY DNA vaccine was injected, immunogenicity increased very significantly. This may have been due to a synergistic effect of 8 P29C3d and Fc IgY on PCAG-sH9.
