Abstract
Objective: Prostate cancer is the second most common cancer in men and a frequent cause of cancer related death in males. Treatments used for this cancer include surgery, radiotherapy, chemotherapy, hormone therapy, immune therapy and targeted therapy (drugs that spe- cifically interfere with cancer cell growth). Coumarin derivatives which are widely distributed in the plants, fungi and bacteria have been extensively investigated as anticoagulation, antiviral, anti-inflammatory, antibacterial and anticancer agents. Coumarins (known as 1, 2-benzopyrones or O-hydroxycinnamic acid-8-lactones) comprise a very large class of phenolic derivatives and consist of fused benzene and α-pyrone rings. Because of the importance of coumarins in the fields of pharmacy, cosmetic and nutrition, these compounds have been widely investigated and many natural and unnatural coumarins have been synthesized. In this research, 8-isopentenyloxy coumarin was first synthesized and then cytotoxic effects of this compound were investigated. The purpose of this study was investigating the cytotoxic and anticancer effects of this compound on PC3 (prostate cancer) and HFF3 (normal human fibroblast) cells. Materials and Methods: PC3 and HFF3 cells were treated with different concentrations of 8-isopentenyloxy coumarin and DMSO as control. After 24, 48 and 72 h cell viability was quantified using spectrophotometer and alamar blue assay. Results: Analysis of cell survival showed that the IC50 values (half inhibitory concentrations) of 8-isopentenyloxy coumarin on PC3 cells were 29.73, 51.14 and 24.57 μg/ml and IC50 values of this compound on HFF3 cells were determined as 35.48, and 17.13 μg/ml. after 24,48 and 72 h of treatments, respectively. Conclusion: Results revealed that 8-isopentenyloxy coumarin could reach its IC50 at 17.13 μg/ml after 72h of treating Hff3 cells. Therefore, Compare ic50 cell lines PC3 and Hff3 at 24, 48 and 72 h, suggesting that this coumarin derivative can be considered as a toxic compound for future in vitro study.
