Abstract

Allergy and immunologic disorders are rising rapidly in prevalence in parallel with high hygiene in developed countries. Several studies based on animal model have demonstrated a negative relationship between clinical asthma and allergy and infection with some helminth species. The immunological basis for this protective effect is not fully clear. Both of them are characterized by elevated of Ig E, eosinophilia and a strong T helper 2 (Th2) responses. Most of helminth infections are generally chronic. Immune response to helminthes is down regulated due to down-regulation of T- and B-cell responses via the induction of regulatory T cells or the anti-inflammatory cytokines IL-10 and TGF-β in the chronic phase of infection. This regulatory response not only regulates immune response against helminth and provides suitable environment for surviving of helmith and host, but also contributions to protection against allergic diseases. In human studies, peripheral T cells from infected patients are nonresponsive to parasite antigens, but responses can be restored by antibodies to IL-10 and TGF-β. It is of great interest to define and characterize specific helminth molecules that have profound immunomodulatory capacities as targets for therapeutic application in the treatment or prophylaxis of allergic manifestations
