Abstract

Background and Aim: Though wild-type microorganisms have been able to successfully produce platinum nanoparticles (PtNPs), but the yield of their biosynthesis is relatively low. As well as the size and composition of the nanoparticles must be controlled. Thus the aim of this study was to compare biosynthesis potential of platinum nanoparticles with a genetically engineered strain Pseudomonas fluorescence os8 and a naturally occurring bacterium Marinobacter adhaerens 2C8. Methods: These bacteria were cultured in LB Broth medium and the cell pellets were separated by centrifugation. For PtNPs biosynthesis, cell-free extract mixed with solution H2ptcl6 (0.01M) at a ratio of 2:1 then incubated for 1 hour. Characteristics of PtNPs were investigated by UV– visible spectroscopy, X-ray diffraction peaks (XRD), Fourier transform infrared spectroscopy (FT-IR), and zeta potential. Results: Preliminary approval biosynthesis of PtNPs was Yellow-brown liquid colloid in solution. Absorption spectra of PtNPs obtained from cell-free extract of p. fluorescence os8 and M. adhaerens 2C8 were recorded at 331 nm and 330 nm, respectively. For both of them XRD results showed 111, 200 and 220 planes of the face-centered cubic. The FT-IR spectra for them clearly showed the presence of primary and secondary amines which may be responsible for the reduction and subsequent stabilization of the resulting extracellular PtNPs. Zeta potential of P. fluorescence os8 and M. adhaerens 2C8 were -21.98 mV and -35.19 mV, respectively. Conclusion: In conclusion, biosynthesis of PtNPs using above bacteria did not show significant difference in case of their characteristics and both strains could synthesis PtNPs in high yield and speed.
