Abstract
Background and Aim: Vibrio fischeri NRRL B-11177 is one of the luminescence bacteria that has special value in biotechnology especially in microbial biosensors construction. Oxidoreductase enzymes of this bacterium have involved in the process of luminescence radiation and the fact that the role of bacterial enzymes in the production of nanoparticles is determined by the type of bacteria is very likely possibility of synthesis of nanoparticles. In this study, we investigated biosynthesis of copper oxide nanoparticle by V. fischeri NRRL B-11177 that has commercial applications in the industry, medical and health sciences, especially as a gas sensor, anti-cancer and anti-microbial agents. Methods: To prepare the nanoparticles, first, 1 ml fresh suspension of the above bacterium (OD600 = 0.1) was added to 98 ml of LB without NaCl. Then 1 ml of copper sulfate solution (Cu(NO3)2.5H2O) at the final concentration of 1 mM was added to the above suspension. After incubation at 30°C and 200 rpm, the change in color from light blue to dark green and eventually brown indicates the formation of copper nanoparticles. The above suspension was centrifuged and the supernatant containing the nanoparticles after filtration was evaluated by ultraviolet spectroscopy (UV-vis), X-ray diffraction spectrometry (XRD), Fourier transform infrared spectroscopy (FTIR) methods. Results: Spectrophotometric observations showed strong and broad peak in 280 nm wavelength and XRD spectra confirmed Monoclinic CuO nanoparticle synthesis with average size of 14 nm. The results of FTIR analysis indicates the presence of regenerative enzymes for the synthesis of nanoparticles. Conclusion: These results approve ability of luminescence bacteria (e.g. V. fischeri) to copper oxide nanoparticles synthesis.
