Introduction and Objectives: in recent years, patients suffering from UTI have shown antibiotic resistance to Escherichia coli (E. coli) strains. In order to develop a more suitable drug, antibiotic cross- reaction should be investigated in multiple geographical areas. This study aims at tracking ESBL resistance genes to E.coli strains in UTI suffers at Ghaem and Emam Reza hospital in Mashhad. Materials and Methods: In this study, 45 E.coli strains extracted from urine samples at Ghaem hospital between years 1393-94, and 60 strains from samples obtained from Ghaem and Emam Reza hospitals between years 1395-96 were investigated. Upon further biochemical examinations, the presence of bla CTX_M , bla TEM and SHV genes were evaluated using specific multiplex polymerase chain reaction (PCR) primers. Results: In the 45 strains studied from Ghaem hospital (years 1393-94), gene bla TEM and bla CTX_M were present in (100%) of the strains while gene bla SHV was detected in 4.4% (2 strains). In the 60 samples investigated from Ghaem and Emam Reza hospitals, gene bla TEM and CTX_M were present in 100% of the samples and gene bla SHV in 6.6% (4 samples). Conclusion: There was no significant difference between the detection of bla TEM gene and bla CTX_M in samples in years 93-94 and 95-96, but there was a significant difference between the detection of SHV gene in years 93-94 and 95-96.
