Background:The advent of regenerative medicine has brought us the opportunity to regenerate, modify and restore organs function.Stem cells,a key resource in regenerative medicine, can generate into one or more specializedcelltypesliketenocytes.Beforeclinicalapplicationofstemcells,theircharacteristicsshouldbedetermined.Thisstudydesignedtoinvestigate weatherdonorhaseffectontenogenicdifferentiationofequine adiposemesenchymalstemcells(ASCs)inresponsetoplateletderivedgrowth-factor(PDGF). Materials and Methods: Equine ASCs derived from three horseswereusedinthisexperiment.ASCsatpassage5were culturedwithbasicculturemediumcontainingPDGF-BB(20 ng/ml),L-Prolinandascorbicacid(AA)at37°Cunder5%CO2 for14days.Thecellsincontrolgroup,wereculturedwithonly basicculturemedium.Attheend,SiriusRedandH&Estainingwereusedtoassesscollagenproduction,asaextracellular matrixproteinfortendon,andcellmorphology. Results:ASCsintreatmentgroupswerechangedtoelongated spindleshapeliketenocytes.TheresultsofSirusRedstaining revealedthattheproductionof collagenhasbeen elevatedin treatment groups comparedwith control groups signi9cantly. Statistical analysis showed thattheproduction of collagenis differentamongASCsderivedfrom3horses. Conclusion:Ourdatarevealedthatcelldifferentiationtoward tenocyteswasdifferentforeachhorses.Inotherwords,donor affecttenogenicdifferentiationpotentialofequineAscs.
