Abstract

Background:Improvementofmusculoskeletaldifferentiation ofmesenchymalstemcells(MSCs)(bothandinvivo) isaninterestingeldsinstemcellresearch.Inordertousestem cells for treatment of orthopedic disorders especially tendon injuries,usingdifferentgrowthfactorsarerecommended.The aimofthis studywasto evaluate differentiation of eq-ASCs inresponsetogrowthdifferentiationgrowthfactor-6(GDF-6). Materials andMethods: Characterized andfrozen eq-ASCs from3horseswerethawedandwereculturedtopassage4(P4). Then,P4cellsweresubjectedtodifferentiationforaperiodof 14daysinaculturemediumcontainingtheGDF-6factor(20 ng/ml)astreatmentgroup.Incontrolgroup,mediumwasthe same astreatmentgroupexceptinducingfactors andGDF-6. After14days,tenogenicdifferentiationwasevaluatedbyH&E and Siriusredstainingforthemorphological assessment and measurement of collagenproduction.Inaddition,the expressionofspecicgenesincludingsclerosis(Scx),collagentype 1(ColIα1)andMohawkwereevaluatedusingqPCRmethod. Results:H&Estainingrevealedastretchingandspindleshape (tenocye-like)ofcellsinthetreatmentgroup.Siriusredstainingdatashowedasignicantincreaseincollagenproductionin treatmentgroupcomparedwithcontrolgroup.BasedonqPCR data, expression of Mkx, Scx and ColI was signicantly increasedinthetreatmentgroupcomparedtocontrolgroup. Conclusion:TheresultsofthisstudyshowedthatGDF-6could inducetenogenicdifferentiationineq-ASCs.
