Abstract
CDDP is one of the most potent antitumor agents; however, it is accompanied by severe side effects, including digestive tract reactions, renal toxicity, bone marrow suppression, and neurotoxicity. Furthermore, long‑term use of CDDP can induce drug resistance. In recent years, much attention has been paid to developing targeted drug delivery systems, which are a promising strategy for increasing therapeutic drug accumulation in cancer cells, while reducing toxicity to normal tissues. Among these, magnetic nanoparticles are attracting more interest. Hence, in the present study, we employed Fe3O4 hydroxyapatite nanoparticles to improve the therapeutic effect of cisplatin (CDDP) in colorectal cancer. So, magnetic hydroxyapatite nanoparticle (mHAP) was fabricated and CDDP was loaded in porous of hydroxyapatite. The synthesized nanocomposite was characterized by a UV–Visible spectrophotometer, FT-IR, and VSM. In vitro, biological experiments revealed that a high efficiency in the cytotoxic effect of CDDP@mHAP in CT26 colorectal cancer, using MTT assay. and Ao/EtBr staining. The IC50 value of treated cells with CDDP (2.84±0.1 µg/ml) was decreased to be 2.34±0.092 µg/ml when the cells were treated with CDDP@mHAP. A pronounced increase in apoptosis was observed in the cells treated with the nanocomposite as compared with that in the cells treated with the CDDP, which is in accordance with the MTT assay results.
