Abstract

In this study, 32 adult male mice were divided into four groups (8 in each group), control (received normal saline) and the three experimental groups (received Crocin 4, 20,100 mg/kg every day) for 6 weeks. At the end of the experiment, blood testosterone level was analyzed by enzyme immunoassay and sperm parameters (count, motility, and viability) were evaluated. Data were analyzed by one-way ANOVA, using SPSS software. After administration of Crocin at a dosage of 100 mg/ kg, the results clearly showed that the number of sperm was significantly reduced compared to the control. In experimental groups (20 and 100 mg/kg), a significant increase in testosterone levels in blood serum compared to the control group was shown. But, in the Crocin group (4 mg/kg), no significant difference in testosterone levels than the control group was shown. Compared with the control group, the average testosterone level of the Crocin group (20 mg/kg) was significantly increased. In conclusion, higher doses (100 mg/kg) of Crocin administration may negatively affect the reproductive system by reducing sperm count, sperm motility, and testicular damage. However, a relatively safe dose of Crocin (20 mg/kg) is required in clinical trials.
