Abstract
Background: Oct4, a POU family transcription factor, is one of the most critical genes during reprogramming and its expression is significantly correlated with stemness state of cells.In this study we suggested a vector which express Green fluorescent protein (GFP) under Oct4 promoter for analysing Oct4 expression in the competent cells. Material & Method: We used a genetic construct which promote GFP under Oct4 gene promoter and transfected the HEK-293T cells using calcium phosphate method.GFP expression was assessed by Fluorescent microscopy 12-24 hrs after transfection. Results: We managed to produce GFP positive,about 40-50%, cells post transfection which highlight the activity of Oct4 gene in the HEK-293T cells. Discussion: Activity of Oct4 in cell lines like HEK is an intriguing issue which precise role of it remain to be clarified. Taken together, Oct4 expression might be used as a reliable marker to reveal the tumoral stage in cancerous cells, and methods that evaluate Oct4 expression of cells could be used as a prognosis marker which correlate with cliniopathological signs of cancers.
