Abstract

Background: Allergic asthma is a chronic inflammatory disease which has affected too many people across the world. Allergic asthma is caused by environmental allergens along with the immune response. Currently, many therapeutic methods are advised for treatment of asthma, but must of them are not effective and clinical symptoms relaps; nowadays novel methods immunotherapy of asthma are being used. This method is based on using of DC as orchestra of immune responses. DCs can polarize Naïve Tcell into different subtypes especially protective T subtypes of T cells. This study aimed at using of DC potent for polarization of immune response to protective one, which can result in production of regulatory Tcell; so that these cells could suppress Th2 immune response and then alleviate the symptom of diseases. Materials and methods: This study has 15 particicpants.10 healthy people and 5 sick people affected by asthma (confirmed by authorized staff of hospital). Their blood samples (contains anticoagulant substances) were collected and their PBMC were isolated and then monocytes separated from lymphocytes. After adding of IL-4 and GM-CSF, monocytes differentiate to DC and at 4th day, antigen extract of Marshallagia marshalli added to culture medium. Then, at 7th day of culture, these DCs cultured by autologous Tcells and finally at 12th day of culture, the supernatants were collected and content of IL-10 and TGF-β were measured by sandwich ELISA. Results: The results of this study indicate that the content of IL-10 and TGF-β in Tcell culture supernatant pulsed by somatic extract of Marshallagia marshalli in asthmatic people increases. Thus, this rising might be resulted from polarizing of Naïve T cell to regulatory T cell. Conclusion: With regards to hygiene hypothesis in which parasitic worms contain regulatory molecule of immune system, it could be possible producing of regulatory DC by antigenic extract of Marshallagia marshalli .
