Abstract

Background and Aim: Amylase is one of the most important industrial enzymes and accounts for about 25% of the total enzymes sales. Cold active amylase is used in the leather, detergents, and food industries. The aim of this study was the effect of different carbon and nitrogen source on amylase production by a newly cold tolerant bacterium isolated from Binaloud Mountain soil. Methods: The amylase activity was measured using Brenfeld method. Influence of different carbon sources including starch, sucrose, maltose, glucose, glycerol and nitrogen sources containing peptone, tryptone, yeast extract, skim milk, ammonium sulphate sources was examined on the production and activity of cold-active amylase enzyme at 20°C during 72h. After optimizing the sources, different concentrations of the best nitrogen and carbon sources were tested by the same method. Results: The results show that 1% of starch with glucose causes the highest amylase activity compared to other carbon sources (125,09 U/ml). However, the nitrogen sources can be decrease enzyme activity. Conclusion: Cold-adapted amylase cold-tolerant bacteria are the good candidate to apply in biotechnological industries.
